Sensitivity Data of AXIOM's Avian Influenza Rapid Tests
[1] The AXIOM Lateral Flow Test requires a one-step performance only. However, to reach a
sufficient high sensitivity, the sample must be prepared for analysis by a three-day incubation in
an embryonated vivid test egg. The test is based on the migration of the colloidal gold-conjugated
immune complex by capillary diffusion to the band-creating test zone.

[2] The AXIOM Dot ELISA is a double antibody sandwich-like assay without pre-arrangements.
It requires a several-step performance, amongst others one with a second monoclonal antibody
conjugated to an enzyme that converts a substrate/dye-mixture.

Special Properties (Advantages) of the AXIOM Dot ELISA:
(A) By the methodology of the AXIOM Dot ELISA [2], the analytical sensitivity is more than threefold
higher than that of the AXIOM Lateral Flow Card [1]. Using an accurately defined, low infectivity of
0.1 hem agglutination (HA) units, the AXIOM Dot ELISA detects about 70 % of the HA-antigen load,
whereas the Lateral Flow Test catches only a bit more than 20 % of it.

(B) Using a geometric dilution series to the basis 2 of an acurately defined H5-HA agent, the AXIOM
Dot ELISA shows an identically high detection limit to AXIOMÕs standard H5-specific antigen-ELISA of 1:512
(in contrast: H5-specific AXIOM Lateral Flow Card 1:4; H5-specific Rapid Test of Competitor1 only 1:4
and of Competitor2 only 1:64).

(C) Using different infectious Influenza A virus isolates for determination of the specific detection limit, AXIOM
H5-HA-specific Dot ELISA shows a limit of 10e5.2 EID50/mL (50 % egg lethal dosis per mL). In contrast,
H5-non-specific Rapid Test of Competitor3 represents only 10e7 EID50/mL.
